SUMMARY or LIST of activities planned in 2006:

Evaluation of a Real-time PCR quantification Method for the risk assessment of the Biocontrol Agent Trichoderma isolate 122F

  • Molecular Characterization of the Trichoderma isolate 122F from the Safecrop collection and of other unknown Trichoderma isolates from Trentino soils from the Safecrop collection.
  • Qualitative PCR assays for the isolate Trichoderma 122F: test for the specificity for two primer sets from ETH.
  • Sensitivity test and extraction efficiency test for primers for qualitative PCR, and comparison with the real-time PCR sensitivity.
  • Quantitative real-time PCR method for the detection and quantification of the Trichoderma isolate 122F: finding of specific primer sets and probe for the isolate, and for the Trichoderma spp.
  • Fast DNA extraction: test on the possibility to perform the PCR directly from the strains from plate medium and from water-drop leaves contaminated with the Trichoderma from greenhouse (strawberries).
  • Quantification of the Trichoderma isolate 122F in soils from two microcosmos and field experiments, in water drops from leaves in open field conditions (grapes and vineyard grass plant leaves) and barbatelle.
  • Real-time PCR tests for the validation of the quantification method following the ENGL (European Network of GMO Laboratories) criteria.
